New markers derived from qualitative and quantitative DNA and RNA have served to extend morphologic interpretations and have thus been increasingly integrated into the final medical diagnosis. DNA and RNA quantifications are widely used in biological and biomedical research. In the era of the Human Genome Project, quantitation of gene expression by cells is of paramount importance in the investigation of gene patterns. Quantitative real time-polymerase chain reaction (RT-PCR) technology has recently reached a level of sensitivity, accuracy and practical ease that supports its use as a routine bioinstrumentation for gene level measurement. It has evolved into the state-of-the-art technique for the quantification of nucleic acids for mutation detection, genotyping and chimerism analysis. The extremely versatile technique of quantitative RT-PCR has had an enormous impact on the life sciences and molecular medicine. In this article, details of this method are reviewed and its haematological usage is examined. Keywords: Polymerase chain reaction, hematology, gene expressionTurkiye Klinikleri J Med Sci 2004, 24:653-660
