The Effect Of Melatonin On Random Pattern Skin Flap Survival In Rat

However, necrosis in distal flap sections resulting from ischemia is a serious problem, increasing the cost of treatment and hospitalization. Free oxygen radicals and increased neutrophil accumulation play an important role in tissue injury and may lead to partial or complete flap necrosis. The aim of this study was to test the effects of melatonin, the chief secretory product of the pineal gland and a highly effective antioxidant, on random pattern skin flaps in rats.
Material and Methods: Herein, to investigate the pharmacological role of melatonin on dorsal skin flap survival, we used concentrations of 0.4, 4 and 40 mg/kg administered intraperitoneally (i.p.) way. The rate of skin flap necrosis was compared between experimental groups by planimetry. To observe the effects of melatonin, levels of malondialdehyde (MDA), nitric oxide (NO), glutathione (GSH), glutathione peroxidase (GSH-Px), and superoxide dismutase (SOD) were measured from extracted skin tissue.
Results: MDA, NO levels were found to be higher in the control group when compared to melatonin groups; GSH, GSH-Px, and SOD enzyme activities were reduced. Melatonin administration to rats reduced MDA and NO levels, increased GSH, GSH-Px, SOD enzyme activities and also reduced the rate of flap necrosis.
Conclusion: These results indicate that pharmacological concentrations of melatonin improve skin flap viability.Keywords: Melatonin, skin, flaps, reactive oxygen radicals, ratsTurkiye Klinikleri J Med Sci 2004, 24:638-644
