Skeletal Muscle Fiber Types

The primitive forms of type 1 and 2 fibers can be discriminated after this time. Indeed, the main histochemical differences occur in the first years after birth.
Skeletal muscle fiber types evidence differences in their morphologic, physiologic and histochemical features. The most reliable method in the discrimination of fiber types has been through histochemical techniques. The most common of these are based upon myofibrillar ATPase and oxidative enzyme reactions. The spectrum of reactions can be extended by modifying the pH during the ATPase staining procedure. In the standard or alkaline ATPase reaction, which is conducted at pH 9.4, two fiber types are seen. Type 1 fibers are pale in staining intensity, whereas Type 2 fibers are dark. If the pH of the incubating solution is brought into the acidic range (pH 4.6), type 2 muscle fibers exhibit two different subtypes. Type 2A fibers are virtually unstained and type 2B fibers are intermediately stained, while type 1 fibers are extremely dark. The oxidative enzyme reaction also reveals two differently stained muscle fibers as type 1 and type 2.
