Protective Effect Of Caffeic Acid Phenethyl Ester On Myocardial Ischemia/reperfusion-induced Histological Alterations

aHistoloji-Embriyoloji AD, bFarmakoloji AD, cAnesteziyoloji ve Reanimasyon AD, İnönü Üniversitesi Tıp Fakültesi, ELAZIĞ Objective: To aim of this study was to investigate the purported protective effect of caffeic acid phenethyl ester (CAPE) against myocardial ischemia/reperfusion.
Material and metods : Twenty-four Wistar rats were randomly divided into three groups: Sham operation, ischemia/reperfusion (I/R) and I/R + CAPE. To produce I/R, the left main coronary artery was occluded for 30 minute, followed by 120 minute reperfusion. The tiobarbituric acid method was used to determine tissue malondialdehyde( MDA) levels as an indicator of lipid peroxidation in the heart. Histological changes were evaluated with ligth microscopy.
Results: I/R was accompained by a significant increase tissue MDA whereas CAPE administration significantly reduced this substrate. I/R induced myocardial damage, which manifested by histopathological evidence of intracelluler vacuolization, reduction in cytoplasmic glycogen granules, neutrophil adhesion to endothelium, mast cell degranulation and coagulation necrosis. These changes induced by I/R were ameliorated with CAPE.
Conclusion: This study shows that CAPE can be regarded as a protective agent action against I/R-induced myocardial damage.Keywords: Ischemia, reperfusion, myocardium, microscopyTurkiye Klinikleri J Med Sci 2004, 24:469-475
