Array Technology: From Chips To Eternity

aTıbbi Biyoloji ve Genetik AD, Kocaeli Üniversitesi Tıp Fakültesi, KOCAELİ Array technology has become an area of great interest in the last decade. “Gene chip” is the commonly used name for this technology. The main characteristic of the methodology is a micro-hybridisation-based assay. A filter, called a chip, is a special kind of membrane in which are spotted several thousand oligonucleotides of cDNA fragments coding for known genes. The resulting hybridisation signal on the chip is analysed by a fluorescent scanner and processed with a special software package.
This technology can be used for many different purposes, most prominently to measure differential gene expression. In addition, it can be used for genotyping and sequencing analysis. Preliminary data indicates that “gene chip” technology may hold high promise and indeed usher in a new age of medicine. It will accelerate our understanding of the complex genetic processes underlying the interaction between microorganisms and the host, with consequent improvements in the diagnosis, treatment, and prevention of infectious diseases. It can also provide new insights into gene function, disease pathophysiology, disease classification and drug development. In this respect, diabetes, hypertension, coronary heart disease, asthma, inflammatory bowel disease, and cancer represent its main targets. In this review, the basic theory of microarray technology and its analytic methods are presented, recently obtained data are reviewed from fields such as oncology, microbiology, as well as in routine diagnostics, and technical issues are discussed in the light of our experience.Keywords: Array technology, routine diagnostics, gene expressionTurkiye Klinikleri J Med Sci 2004, 24:534-540
