Effect Of Transforming Growth Factor-alpha Microemulsion Form On Wound Healing In Peptic Ulcer Models With And Without Ischemia And Relationship Between Transforming Growth Factor-alpha And And And Malondialdehyde, Glutathione And Nitric Oxide Levels

bFarmasötik Teknoloji AD, Gazi Üniversitesi Eczacılık Fakültesi, ANKARA Objective: Transforming Growth Factor-Alpha (TGF-α) is affected by gastric pH. Drug formulae incorporating this factor may serve to increase its effects on ulcer healing. This study was planned to investigate the effects of microemulsion forms of TGF-α on healing rates of peptic ulcer formed by ischemia and/or aspirin (ASA), as well as to review the relationship between ulcer, TGF-α and oxidative processes.
Material and Methods: Eighty-five male Wistar albino rats (200 ± 25 g) were used in this study. Animals were divided into 10 groups. After ulcer induction by ASA and/or ischemia in the treatment and control groups, ulcer areas were detected by planimetry as mm2. Determinations of gastric mucus amounts, malondialdehyde and glutathione levels were measured by spectrophotometry. Total nitric oxide levels were detected by VCI3 + Griess reactants and ELISA reader. Results were evaluated with ANOVA and Mann Whitney U Tests, and a criterion significance level of p< 0.05 was used.
Results: Our results showed that TGF-α microemulsion treatment decreased ulcer areas and malondialdehyde levels, and increased mucus and total nitric oxide levels compared to non-treatment groups (p< 0.05). Glutathione levels were not changed. Ulcer areas, malondialdehide and total nitric oxide levels in the ischemic groups were higher than the other groups, whereas glutathione levels were not (p< 0.05).
Conclusion: The microemulsion form of TGF-α is effective in increasing the healing rate of ulcers formed by ASA and ASA + ischemia.
