Endotelin peptides are local peptide hormones produced by vascular endothelial cells anda re the most potent vasoconstrictors yet identified. The vasoconstrictor properties of these peptides are powerfull and long-lasting. THey were first discovered in cultured endothelial cells but endothelin expression has been found in many other tissues such as kidney and brain. These are peptides with 21 amino acids formed by hydrolytic cleveage of a larger peptide, big endotheline. Spesific high affinity binding sites for the endothelins are widely distributed in the vasculature, heart, kidneys, lung, adrenal glands and brain. Literature now abounds with reports on actions of the endothelins in vitro and in vivo. There are increasing evidences that endothelins play a role in the regulation of vascular tone and blood pressure and in the pathogenesis of cardiovascular, renal and respiratory disease. IN this review the authors have summarized the literature data concerning this subject.
