Superoxide Dismutase, Glutathione, Vitamin C, Total Antioxidant And Total Thiol Levels In Hydatid Cysts

aBiyokimya AD, Ondokuz Mayıs Üniversitesi Tıp Fakültesi, SAMSUN Objective: Cystic echinococcosıs (CE) has been investigated for many biochemical aspects. However, the relationship between growing CE and antioxidants, especially with respect to known hydatid cyst anti-oxidant mechanisms against host oxidants, has not been sufficiently clarified.
Material and Methods: In this study, the levels of superoxide dismutase (SOD) in fertile (sheep and cattle, liver and lung) and infertile (cattle, liver and lung) CE membranes, and total thiol and reduced glutathione levels in both membranes and protoscoleces, as well as total antioxi-dant status, total thiol, reduced glutathione, vitamin C levels and SOD activity in fertile and infertile hydatid cyst fluids were determined.
Results: Total antioxidant, total thiol, reduced glutathione and vitamine C levels in fertile cyst fluids (liver and lung; 1.42 ± 0.52, 1.21 ± 0.09 mmol/L; 31.6 ± 3.70, 28.0 ± 3.90 μmol/L; 7.21 ± 1.03, 6.96 ± 0.80 μmol/L; 0.43 ± 0.08, 0.32 ± 0.04 mg/dL, respectively) were higher than in cattle infertile hydatid cyst fluids (liver and lung; 0.80 ± 0.13, 0.87 ± 0.13 mmol/L; 14.70 ± 1.58, 16.90 ± 3.82 μmol/L; 3.03 ± 0.30, 3.02 ± 0.18 μmol/L; 0.08 ± 0.01, 0.09 ± 0.01 mg/dL, respectively) (P< 0.05). SOD activity in sheep fertile cyst fluids (liver and lung; 1.35 ± 0.99, 1.12 ± 0.52 U/mL) and its cyst membranes (liver and lung; 1.76 ± 0.66, 1.50 ± 0.49 U/mg tissue protein/min.) were found to be significantly higher compared to cattle fertile cyst fluids (liver and lung; 0.47 ± 0.30, 0.50 ± 0.24 U/mL) and its cyst membranes (liver and lung; 0.55 ± 0.16, 0.36 ± 0.28 U/mg tissue protein/min.), as well as in cattle infertile cyst membranes (liver and lung; 0.15 ± 0.05, 0.12 ± 0.02 U/mg tissue protein/min) (P< 0.05). However, in cattle, infertile hydatid cyst fluids could not be measured for SOD activity.
Conclusion: These results show that the higher levels of total antioxidant status, total thiol, reduced glutathione and vitamin C in fertile hydatid cyst fluids with parasite larvae, in comparison with those in infertile hydatid cyst fluids, as well as the higher levels of SOD activity in fer-tile hydatid cyst membranes in comparison with those of infertile hy-datid cyst membranes, may be evidence that these antioxidants might be implicated in the antioxidant defence systems of parasites, in par-ticular in fertile hydatid cyst membranes and parasite larvae.Keywords: Antioxidants, fertility, hydatid cystsTurkiye Klinikleri J Med Sci 2004, 24:213-218
