Obtaining Of Ubiquitin Activating Enzyme And Kinetic Features

Abnormal proteins synthesized within cells are degraded by snown two mechanisms. One is the degradation of long-lived proteins found or extracellular membranes by ATP dependent lisosomal system second mechanism, known ubiquitin system functions to degradade abnormal short-lived proteins. The enzyme that is responsible for ubiquitin activation were purified from plasenta using affinity chromatography. The enzyme that is responsible for ubiquitin activation were purified from plasenta using affinity chromatography. The enzyme purity were checked by SDS-PAGE and kinetic features were examined. It has been found that ethilmaleimid (C6H7NO2), EDTA and Hg (NO3)2 were inhibitor of the enzyme optimum pH of the enzyme was 7 and optimum temperature was 35oC.
