Leptin, Insulin And Diabetes

** Doç.Dr., GATA Tıp Fakültesi Analitik Toksikoloji BD, ANKARA Leptin is a hormone produced by adipocytes and is thought to play a key role in the control of body weight. The discovery of leptin raised the hopes of obese population. Leptin inhibits feeding, stimulates thermogenesis, and decreases body weight through distinct receptors in target organs, which controls food intake and energy metabolism. Its absence experimentally and clinically produces obesity. Adiposity and insulin resistance are important risk factor in diabetes mellitus. Obese individuals characteristically manifest insulin resistance and hyperinsulinemia, which predispose to glucose intolerance, diabetes, and cardiovascular disease. In humans, insulin stimulates leptin production in adipocytes. Recent studies suggest a complex relationship between leptin and insulin resistance. Leptin has been shown to increase insulin sensitivity and improve glucose metabolism. It normalizes the hyperglycemia and hyperinsulinemia in ob/ob mice at doses that do not decrease weight.
