Effects Of Pentoxifylline Application On Tnf-a, Icam-1 Levels And On The Histology Of Liver In The Rabbits Exposed To Experimental Sepsis Model

***** Doç.Dr., Fırat Üniversitesi Tıp Fakültesi Patoloji AD, ELAZIĞ Purgose: Inflammatory cytokines especially tumour necrosis factor-alfa (TNF-α) are released excessively and uncontrolled in sepsis. Intercellular adhesion molecule-1 (ICAM-1) which TNF-α provides it to be defined at endothelial cells and circulatory release is increased by it is held accountable from liver damage and dysfunction. Treatments included towards the modulation of inflammatory response is being used at sepsis. At this study the correlation between liver dysfunction, mortality and circulating ICAM-1 concentrations in sepsis during pentoxifylline application was searched.
Material and Methods: In the study, the rabbits were separated into 3 random groups. One hour after cecal ligation and puncture (CLP) at rabbits 50 mg/kg IV pentoxifylline to pentoxifylline group and serum saline to sepsis group was given. Control group received only laparotomy. Physical examination was noted 1 day before CLP and daily after operation. Biochemical studies were done 1 week before CLP and for three periods after operation. At the end of experiment tissue MDA levels at liver were measured and histological evaluation was done. During the experiment dying animals were noted and survive ration were calculated.
Results: At pentoxifylline group TNF-α, sICAM-1, lactate and tissue MDA levels were similar to sepsis group. Increase at AST and ALT levels were significant at sepsis group than pentoxifylline group. Histological differences of liver preparations were a little significant than control group. Survive ratios were not different at pentoxifylline group from sepsis group.
Conclusion: As results, no evident effect of pentoxifylline on modulation of inflammatory response and mortality was determined, but it has been that it decreased the histologic differences at liver and prevented liver dysfunction.Keywords: Pentoxifylline, Tumor Necrosis Factor, Intercellular Adhesion Molecule-1, Liver Diseases, mortalityTurkiye Klinikleri J Med Sci 2003, 23:135-140
