Morphological Changes Of Rat Endocrine Pancreas In Experimental Diabetes

**İnönü Üniversitesi Tıp Fakültesi, Farmakoloji AD, Malatya Purpose: Diabetes mellitus is a metabolical disease which developes when the insulin hormone released from b cell is insufficent or non-existent.The purpose of this study was to examine and evaluate the light microscopic changes in the morphology of the islets of Langerhans in the rat following streptozotocin (STZ) and fructose- induced experimental diabetes. Material and metods : Fifteen male Sprague- Dawley adult rats were divided into three groups. One has fed by nornal diet and tap water (control group, n=5), second has given one dose of 45 mg/kg STZ intraperitoneally and fed by normal rat diet (STZ group, n=5) and tap water, third has fed by normal rat diet and tap water containing 10 % D-Fructose (fructose group, n=5) for 8 weeks. At the end 8 weeks, their pancreas was examined by light microscope.Results: Plasma- glucose level was 445.8 ± 73.8 mg/dl in STZ group while 158.4 ± 28.8 mg/dl in fructose groups (p<0.05). In both STZ and fructose groups pathologic evidence were not present in exocrine pancreas. Islets structure was distrupted in STZ group. Pancreatic islets were smaller in STZ group when compared with the fructose and control groups. Secretory granules of b cells were found decreased both STZ and fructose groups with Aldehyde -fuchsin (A-F). Conclusion: This study showed that the shape and border of the pancreatic islets were disrupted in STZ group. Whereas islet structure in fructose group was closer to the control .
