The Effect Of Anti Thyroid Drugs In The Rat Parafollicular (c) Cells

*** Prof.Dr., Fırat Üniversitesi Tıp Fakültesi, Histoloji ve Embriyoloji AD, ELAZIĞ Purpose: The aim of the study was to investigate the effect of anti thyroid drug (propyl thiouracil) on the thyroid follicular epithelial and C cells. Material and Method: In this experimentally study, hypothyroidism was induced by 10mg/kg 6-n-propyl-2-thiouracil (Sigma) by injecting to adult male rats intraperitoneally. The rats were separated into three groups (4 rats/group). One group of animals that did not receive the propyl thiouracil was considered as the control group. Second group of animals were treated with propyl thiouracil for 15 days. Third group of animals, after 15 days of treatment were allowed free access to tap water without propyl thiouracil with survival times of 15 days. All animals were killed under ether anaesthesia by decapitation and thyroid glands were removed. The material cut into small pieces and fixed in 2.5% glutaraldehyde in 0.1 M phosphate buffer. Samples were postfixed with 1% osmium tetraoxide and embedded in Araldit.Results: We observed that follicular lumens were narrow, irregular and depleted of colloid and columnar epithelial and C cells displaying many more mitotic figures in the thyroid glands of 2 groups. The mitotic activation in the epithelial cells was higher than C cells. At 15 days after the withdrawal of propyl thiouracil treatment, the follicular lumens became wider, more regular and filled with colloid. Mitotic figures were not visible in the follicular epithelium and C cells. C cells were hypertrophic and hyperplasic. Ultrastructurally the C cell cytoplasm had a dilated rough endoplasmic reticulum cisternae and variable size secretory granules.Concluision: Our results indicate that after cessation of propyl thiouracyl in induced hypothyroidism thyroid follicular cell return to normal but C cells do not.Keywords: Propylthiouracil, Microscopy, Thyroid GlandTurkiye Klinikleri J Med Sci 2002, 22:497-501
