The Distribution Of Fibronectin And Its Izoforms In Fetal Membranes: A Biochemical Analysis

Fibronectin isoforms (Fni) seem to play an important role during trophoblast invasion. Besides, there are speculations that small fibronectin like molecules carrying antigenic epitope of “i” blood precursor supress the immune system of the mother. The aim of the present study was to analyze the differences between the other types of matrices produced by maternal and fetal tissues and the tumor-like matrix produced by trophoblast. The main concern was focused on the distribution of Fni in the extracellular matrices. On the other hand, it was also aimed to isolate and characterize small fibronectin like molecules carrying antigenic epitope of “i” blood precursor. Tissue samples were collected from different sites of human term placenta by using immunohistochemistry the expression of Fni (general, cellular and oncofetal) were determined. Besides, matrix components of these tissues were extracted with a multi-step extraction protocol. The presence of Fni were demonstrated by SDS-PAGE and Western blots. The signals from immunoreactivities and enzymatic activities were detected by scanalytics system. The extractions from fetal membranes were applied on gelatine agarose and affinity columns to isolate and purify the small fibronectin like molecules carrying antigenic epitope of “i” blood precursor. In nearly all tissues reactivities and activities for Fni were determined. These molecules were co-extracted from all tissues in variable degrees. Strongest reactivities for cellular and oncofetal fibronectin was found, in particular, in areas, where invasive fetal trophoblast cells invade maternal tissues. The results obtained during the isolation and purification of small fibronectin like molecules carrying antigenic epitope of “i” blood precursor were discussed regarding to their specificity and their relevance reported in the literature.Keywords: Placenta, Trophoblasts, Extracellular Matrix, FibronectinsTurkiye Klinikleri J Med Sci 2002, 22:449-460
