Effects Of Deoxycorticosteron, Ouabain And Combinations Of Deoxycorticosteron And Ouabain On Blood Pressure In Rats

In order to test the hypothesis that ouabain may amplify the hypertensive action of mineralocorticoids, we studied effects of administration of ouabain and DOC alone and in combination on blood pressure for up to five weeks in uninephrectomized rats drinking 0.9 % NaCl. Normotensive, male Wistar rats were unilaterally nephrectomized. After a week of recovery from surgery, graded doses of DOC (5 mg, 10 mg), ouabain (1 mg, 2 mg) and a combination of DOC (5 mg) and graded doses of ouabain (0.05 mg, 0.1 mg, 0.25 mg ve 1 mg) were injected into rats weekly for 5 weeks. DOC and ouabain were dissolved in 1 ml olive oil and injected s.c. Rats in control groups received 1ml olive-oil only. All rats drank a 0.9 % NaCl solution, their systolic blood pressures were monitored weekly. It was observed that blood pressure of rats given 5 mg DOC or ouabain alone was not significantly different compared with control rats. However, the blood pressures of rats given the larger dose DOC (10 mg) and the combinations of DOC (5 mg) and ouabain (0.25, 1 mg) were markedly higher than control rats. The present results show that long-term administration ouabain does not produce hypertension, whereas a combination of small dose DOC and ouabain produce hypertension like large dose DOC.Keywords: Blood pressure, Desoxycorticosterone, OuabainTurkiye Klinikleri J Med Sci 2002, 22:505-509
