The Effects Of Oxidative Damage Induced By Chlorpryfos- Ethyl On The Rat Liver

****Doç.Dr., Süleyman Demirel Üniversitesi, Tıp Fakültesi Biyokimya AD, ISPARTA Purpose: In this study, we aimed to evaluate the toxic effects of chlorpryfosethyl, histologically and biochemically in the rat liver. Materials and Methods: In this study, 40 Wistar albino rats were used. Twenty of them were belonging to control group. Chlorpryfosethyl was administered in two equal doses (41 mg/kg body weight) by gastric intubation to the experimental group for two days. The experiment was ended after three hours from the last application. The tissue samples were taken by using appropriate techniques for histological and biochemical examination. Light microscopic techniques were used to the tissue samples for histological examination. In addition, thiobarbituric acid reactive substances (TBARS) and antioxidant potential (A-OP) levels and glutathione peroxidase (GSH-Px), catalase (CAT), superoxide dismutase (SOD) activities were determined in the tissue homogenates.Results: In tissue homogenates, thiobarbituric acid reactive substances (TBARS) were found to be increased in Group II, when compared to Group I (p<0.05). But, antioxidant potential (AOP) was found lower in Group II than in Group I (p<0.05). Glutathione peroxidase and catalase activities were decreased in Group II (p<0.05), when compared to Group I. Superoxide dismutase activities were lower in Group II than Group I, but it was not significant statistically (p>0.05).In the liver tissues of the rats exposed to chlorpryfos-ethyl, the significant structural changes we observed were as follows; biliary proliferation, mononuclear cell infiltration around some central veins, sinusoidal dilatation and congestion. In addition, connective tissue proliferation was observed in the some portal area. Conclusion: In conclusion, we showed that chlorpryfos-ethyl impairs the antioxidant defence system and reduces the antioxidant defense potential in liver tissue.Keywords: Mitochondrial oxidative damage endonuclease, Rats, Liver, HistologyTurkiye Klinikleri J Med Sci 2002, 22:50-55
