Toxic Effects And Elimination Of Ochratoxin A

** Prof.Dr., Süleyman Demirel Üniversitesi Tıbbi Biyoloji ve Genetik AD, ISPARTA Various studies indicate that the mycotoxin Ochratoxin A (OTA) is carcinogenic, genotoxic, teratogenic, immunotoxic and nephrotoxic. The sequence of effects underlines the importance of this substance such as DNA breakage, inhibition of protein biosynthesis and gluconeogenesis, lipid peroxidation, disruption of oxidative phosphorylation in mitochondria, inhibition of blood clotting and apoptosis. Part of the toxin is subjected to a hepatic first-pass elimination and is removed by the bile before it can enter the systemic blood circulation. Another important organ of elimination is the kidney. OTA can be reabsorbed in all nephron segments, this process may lead to accumulation of the toxin in renal tissue and to the increase of its toxicity. The carcinogenicity of OTA was evaluated in three strains of mice and in one strain of rat; the kidney, and in particular the tubular epithelial cells, was the major target organ for OTA-induced lesions. OTA induced atypical hyperplasia, cystadenomas and carcinomas of the renal tubular cells in male ddY and DDD mice, as were neoplastic nodules and hepatocyte tumours of the liver. In male and female F344 rats, OTA induced nonneoplastic (degenaration, karyomegaly, proliferation, cytoplasmic alteration, hyperplasia) and neoplastic effects (adenomas and carcinomas with metastases) in the kidneys; the incidance of fibroadenomas of the mammary glands was also increased in female rats.
