Immunohistochemical Determination Of Desmin And Vimentin In The Rat Liver Tissue At Hypervitaminosis A

*** Araş.Gör.Dr., Fırat Üniversitesi Tıp Fakültesi Patoloji AD, ELAZIĞ Purpose: The aim of this study was to determine desmin and vimentin immunohistochemically in liver tissue of rats exposed to high dose of vitamin A. Material and Method: Adult Wistar male rats, weighing 200-250 g, were treated with vitamin A at 20 mg/kg and 40 mg/kg oral gavage, for 15 days. The control groups were fed normally. At the end of the study paraffin blocks were prepared from the liver tissue. Avidin-Biotine Immunoperoxidase technique was applied to 5mm thick paraffin sections. Monoclonal Anti-Human Desmin, D33- Dako, and Mouse Anti-Vimentin, V9- Dako primer antibody were used. Results: Desmin (+) immun staining was not observed in both experimental and control groups. In experimental groups, vimentin (+) immun staining increased compared to the control group, particularly in Kupffer cells. The activation of Kupffer cells in the group with 40 mg/kg vitamin A was significant and it was also observed that cell infiltration areas was stained intensively with vimentin.Conclusion: The high dose of vitamin A caused to the activation of Kupffer cells in the liver tissue.
