Immunoreactivity Of Matrix Metalloproteinases In Human Full-term Pla-centa And Their Possible Roles During Delivery

Matrix metalloproteinases (MMP) that are present in this matrix, seem to play an important role during trophoblast invasion. The aim of the present study was to analyze the differences between the other types of matrices produced by maternal and fetal tissues and the tumor-like matrix produced by trophoblast. The main concern was focused on the distribution of MMP in the extracellular matrices. Tissue samples were collected from different sites of human term placenta by using biochemistry methods, the expression of MMP were determined. Besides, matrix components of these tissues were extracted with a multistep extraction protocol. The presence of activities of MMP (MMP-2, -3, -9) were demonstrated by immunohistochemistry and gelatine-substrated SDS-PAGE (zymography). The signals from immunoreactivities and enzymatic activities were detected by scananalytics system. In nearly all tissues reactivities and activities for MMPases were determined. These molecules were co-extracted from all tissues in variable degrees. Strongest reactivities for strongest enzymatic activity for MMP-2 was found, in particular, in areas where invasive fetal trophoblast cells invade maternal tissues. MMP-9 was found to have the highest enzymatic activity in the deepest part of human placenta at the contact region of fetal and maternal part, suggesting the importance of MMP-9 in the separation of placenta from uterine wall during the labor.Keywords: Placenta, Trophoblasts, Pregnancy, Extracellular matrix, Matrix MetalloproteinasesTurkiye Klinikleri J Med Sci 2002, 22:354-361
