Obesity, Food Intake, Control Of Body Weight And Leptin

Body weight is preserved by the balance between gained energy from food and used energy. Lipid cells are active cells that shows changes in volume continuously and in lipid cells excess energy is deposited and energy deficit is compensated. While hormons as insulin, adipsin and cortison causes lipogenic effect in adiposit tiroxin, growth hormone, adrenaline and noradrenaline causes lipolytic effect. Adiposit culture studies show that adiposit have endocrine functions. Certain proteins which were synthetisized by adiposit are: apolipoprotein E, angiotensinogen, IGF-1, adipsin, TNF-a and leptin.
Food intake is regulated by reciprocal working of satiety and hunger centers. Hypothalamus acts through effecting basal metabolism, controlling food intake, changing energy usage and physical activity.
There are short term and long term regulations of food intake. Short term regulation is controlled by direct stimulatory effect of food in the gastrointestinal tract and indirectly by hormones such as CCK which are secreted from the gastrointestinal tract. NPY containing neurons in the hypothalamus has a major role in longterm control of food intake. Many transmitters and hormones which effect these neurons effects food intake. It appears that leptin is the most important and actual one among these. Leptin is a protein composed of 167 a.a. and it is secreted from lipid cells, is carried to the CNS by blood and shows its effect through its receptor. It decreases food intake by supressing NPY synthesis in the hypothalamus. Leptin is produced by ob-gen by encoding to mRNA in the lipid cells. Plasma leptin amount increases as the numbers and volumes of lipid cells increases Leptin decreases body lipid content by decreasing plasma glucose and insulin levels and increasing metabolic rate and physical activity.
So it seems that it will take place in the treatment of obesity.
