Interaction Between Alpha-adrenergic Receptor Agonistsand Antagonists In Canine Isolated Saphenous Vein

In this in vitro study, a-adrenergic receptor agonists phenylephrine (FE), noradrenaline (NA), clonidine, UK 14303 and B-HT 933 and a-adrenergic receptor antagonists, prazosin, yohimbine and fluparoxan have been used to investigate alpha-adrenergic receptor subtypes in canine isolated saphenous vein.
The strips have been suspended in 25 ml organ bath containing Krebs-Henseleit solution which was maintained at a constant temperature of 37oC and bubbled with a gas mixture containing 95% O2 and 5% CO2. The contractile responses of cumulative concentrations of a-adrenergic receptor agonists have been obtained. Then, the inhibitory potencies of a-adrenergic receptor antagonists applied in two different concentrations have been investigated. All responses have been recorded isotonically on a smoked paper.
It has been concluded that five a-adrenergic receptor agonists used in canine saphenous vein caused dose-dependent contraction and their affinities are UK 14304 > clonidine = B-HT 933 = FE > NA, respectively. It has been established that in canine saphenous vein clonidine-yohimbine, UK 14304-fluparoxan, B-HT 933-yohimbine and FE-prazosin interaction suited to competitive and contrarily NA-prazosin interaction suited to non-competitive antagonism type.
In conclusion it has been found out that postsynaptic a-adrenergic receptors in canine saphenous vein suited to a1- and a2-adrenergic receptor subtypes.
