The Effects Of Chronic Manganese Chloride Injection On Plasma Erythrocyte, Brain Manganese Brain Malondialdehid Levels And Histology

It is known as an free oxygen radical scavencer, but when it is present in excess quantities in tissues it may be toxic. These toxic effects are seen in deep brain tissues especially in pyramidal neurons and known to induce a parkinsonian state. The use of Manganese (Mn) and its compounds, in industry, generates adverse health effects in workers who are exposed to Mn. The toxicity of manganese in brain has not been fully understood. In the present study we aimed to investigate the effects of high dose (30mg/kg/day) and chronic Mn application on brain tissue. In the experiments, 20 Wistar rats were used. Ten of the rats were injected MnCl2 as a dosage of 30 mg/kg/day for 50 days and kept as experimental (group II). The remainder of the10 rats were injected serum physiologic at the same time and same amount as in experimental group and kept as control group (group I). Rats were fed with granulated food and water at libitum. At the end of the experimental period rats were fasted 12 h, they were weighted. Under light anesthesia blood samples were collected intracardically in tubes(washed with deionised water before) having heparin for the routine measurements and plasma and erythrocyte Mn levels. Than the brain were removed and put into three pieces. One was fixed in 10% formaldehyde for the histologic investigation, from the remainder two parts, brain tissue Mn (cerebral cortex and cerebellum) and malondialdehid (MDA) levels were measured. The statistical analysis were evaluated by Mann Whitney U and Kruscal Wallis tests. There were not considerable difference between initial and final weights of rats and the brain MDA levels. The manganese levels in plasma, erythrocyte, and brain tissue increased significantly (p<0.001). In the histological investigation of brain tissue, lipofucsin granules were observed in pyramidal neuron cytoplasms showing an inganese may lead to the severe damages in brain tissue and neuronal dysfunctions later in time, because of the defense mechanisms of the body such as less absorption and much excretion in biliary system even the antioxidative effect of Manganese itself.Keywords: Manganese, Central Nervous System,
