The Effect Of Castration On The Adrenomedullinin The Rat Penise, Prostate And Seminal Vesicles

In this study we evaluated the ADM concentration in the penis, prostate and seminale vesicles (SV) of castrated rats. A total of 24 male Spraque -Dawley rats were involved in the study. Rats were divided into three equal group: Group-I control, Group-II orchiectomised rats and Group-III orchiectomized plus testosterone treatment group. After 60 days of bilateral castration rats were sacrified and penis, prostate and seminal vesicles were removed for ADM detection. ADM levels of homogenized tissues were determined using HPLC and results were compared with Mann-Whitney U test. Mean ADM concentration in the penis, prostate and seminal vesicles was found as 461.62±25.56; 488.05±39.37; 390.52±19.66; 178.68±11.80; 163.16±18.16; 103.36±10.64 and 450.14±20.30; 475.10±34.07; 385.56±21.06 pm/L in control group, orchiectomy group and testosterone received group, respectively. Mean ADM concentration was lower in orchiectomized group than in control and testosterone received groups (p<0.05).
The lower concentration of ADM in the testosterone dependent organs after castration reveals that testosterone is important in these organs in terms of ADM expression. To determine the clinical significance of this finding in the erection physiology, voiding dysfunction due to prostatic enlargement and male infertility further studies are needed.Keywords: adrenomedullin, orchiectomy, penis,
