Analysis Of Cardiac Effects Of Short Term And High Dose Growh Hormone Treatment In Rat

Heart is the most affected organ by applications of GH. Recent years, it has been reported that short-term application of GH might be useful for the treatment of many cardiac disease. Unfortunately, the effect of GH applications on the cardiac tissue is not known completely yet. The, aim of this study is the investigation of the alterations of short-term and high dose of GH application on the cardiac muscle cell and vessels.
Material and Methods: 3 months aged 30 wistar rat are divided into equal 3 groups. First group was treated with 0.3 mg/kg/day GH, and second group was used as a control, which was treatment of equal volume of serum physiologic(SP), applied by intraepidermal single dose-per day for ten days. Nothing was applied to third group(C). Animals were sacrificed at the end of the experiment and their hearts were removed. Tissues were processed for histochemistry, immunohistochemistry and electron microscopy.
Results: There was no difference of the volume of the coronary arteries (C: 9.7±1.2, SP: 9.4±0.9, GH: 11.6±1.1 cm3), the endothelial diameter of the coronary vessels (C:0.64±0.15, SP:0.60±0.17, GH: 0.73±0.014 mm), the endothelial thickness of the coronary arteries (C: 0.004±0.001, SP: 0.004±0.001, GH: 0.005±0.001 mm) and the thickness of the coronary arteries (C: 0.015±0.004, SP: 0.016±0.003, GH: 0.014±0.004 mm) among three groups for tissue, vessels and basal membrane architecture of heart at light microscopic level. However showed that there was endothelial hypertrophy (C: 9.7±6.5, GH: 26.3±9.8 mm, p< 0.0001), occlusion (C: 79.7±43.4, GH: 59.4±21.8 mm, p< 0.016), and Conclusion: These ultrastructural alterations for the effect of GH on the capillaries were seen starting from tenth day. Therefore, it was concluded that patients who were treated with GH should be carefully followed for a long time period because of the possible complications.Keywords: Growth hormone, Heart, Endothelium,
